384 WELLED GELS
 384

 - 4 x 96 wells in a 12 x 8 format
 - 4 x 12 wells as markers, outside the arrays
 - Well size 1.5 x 1.5 x 1.5 mm
 - Well volume 3.375ul
 - Lane length 10mm

The 384-well MADGE system is used in conjunction with 384-well PCR for very high throughput genotyping. The same angle as stretch-MADGE (78.7º) is used, allowing four arrays of 96 samples to be run in tracks that are 1mm wide by 11.5mm long. Four arrays are loaded on one gel and
run for 8 to 10 minutes at 150V - this allows very rapid data acquisition from PCR genotyping assays.

The wells are loaded using a 96-pin passive replicator, which transfers about 2µl of PCR product to each well. Loading is "semi-dry", so electrophoresis must be in a semi-dry system. This can be a custom-built electophoresis chamber in which electrodes make direct contact with the gel, or a standard tank, filled below the level of the gel with buffer and fitted with wicks which complete the circuit (Figure 4). Four transfers load one 384-well gel, however it is normal to double-load each well to increase band intensity
