The 96-well MADGE system is used in conjunction with any 96-well PCR for high throughput genotyping. The angle MADGE (78.7º) is used, allowing 96 samples to be run in tracks that are 2mm wide by 27mm long. The arrays are loaded on a single gel  and electrophoresed either in standard wet system or semi dry for 20-30 minutes or 8 to 10 minutes at 150V, respectively - this allows very rapid data acquisition from PCR genotyping assays.                                                                                                                                             back to 'concept'

The wells are loaded using either a standard multi channel pipettes or a 96-pin passive replicator, RepMate, which transfers about 2µl of PCR product to each well. Loading with the pipette is carried out in the electrophoresis system, loading under buffer or in the  "semi-dry" system, on the bench with the RepMate, into dry wells.

Madge Bio offer a standard wet gel tank,adapted for MADGE applications to overcome the "edge effects" seen in off the shelf models. A custom-built electophoresis chamber, in which electrodes make direct contact with the gel has been developed by the Southampton laboratory.

Band intensity can be increased by double loading the gel with the RepMate - ie 2 x 2ul.
